目的探讨缺血性脑卒中患者血小板活化标记物血小板糖蛋白Ⅵ(glycoprotein,GPⅥ)、P-选择素(CD62P)的表达水平及葛根素治疗对其的影响。方法收集2009年1-6月佛山市第三人民医院首发的缺血性脑卒中住院患者50例和同期体检的正常对照50例作为研究对象;采用流式细胞仪(FCM)检测两组GPⅥ和CD62P的表达水平并观察患者葛根素治疗前后GPⅥ和CD62P的表达变化;应用双抗体夹心ELISA法检测两组血浆中可溶性GPⅥ水平;应用直线相关分析统计血小板膜GPⅥ与CD62P、血小板膜GPⅥ与可溶性GPⅥ之间的相关性。结果缺血性脑卒中患者(病例组)血小板膜GPⅥ和CD62P表达增加,并且明显高于正常对照组(P<0.01);葛根素治疗后血小板膜GPⅥ、CD62P表达有明显下降(P<0.05),但仍高于正常对照组(P<0.05);缺血性脑卒中患者血小板膜GPⅥ与CD62P之间呈显著直线正相关(r=0.784,P<0.01);病例组可溶性GPⅥ水平明显低于正常对照组(P<0.01);血小板膜GPⅥ与血浆可溶性GPⅥ之间呈直线负相关(r=-0.828,P<0.01)。结论 GPⅥ和CD62P均可作为缺血性脑卒中血小板活化的标志物,而且二者具有同样的敏感性;GPⅥ、CD62P检测对于指导用药有临床价值;血浆可溶性GPⅥ亦可作为血小板活化的评估指标。
Objective To investigate the expression of platelet glycoprotein (GPVI) and P-selectin (CD62P) in patients with ischemic stroke and the effect of puerarin on it. Methods Fifty hospitalized patients with ischemic stroke in Foshan Third People’s Hospital from January to June in 2009 and 50 normal controls in the same period were enrolled in this study. Flow cytometry (FCM) CD62P and the expression of GPVI and CD62P before and after puerarin treatment. The level of soluble GPVI in plasma was detected by double antibody sandwich ELISA. The linear correlation analysis was used to detect the expression of platelet membrane GPVI and CD62P, platelet membrane GPVI and soluble GPVI The correlation between. Results The expression of platelet membrane GPVI and CD62P in ischemic stroke patients (case group) increased significantly (P <0.01), while the expression of GPVI and CD62P in platelet membrane decreased significantly after treatment with puerarin (P0.05) , But still higher than the normal control group (P <0.05). There was a significant linear positive correlation between GPVI and CD62P in ischemic stroke patients (r = 0.784, P <0.01) (P <0.01). There was a linear negative correlation between platelet membrane GP Ⅵ and plasma soluble GP Ⅵ (r = -0.828, P <0.01). Conclusions Both GPVI and CD62P can be used as markers of platelet activation in ischemic stroke, and both of them have the same sensitivity. The detection of GPVI and CD62P is of clinical value in guiding the administration of drugs. Plasma soluble GPVI can also be used as an index of platelet activation.
