目的 研究红毛五加多糖对胃癌细胞的抑制作用及其作用机制。方法 检测细胞群体倍增时间以了解该多糖对胃癌细胞增殖的影响 ;使用流式细胞仪检测该多糖对抑癌基因 p5 3、Fas及Fas L蛋白表达的影响。 结果 红毛五加多糖 3个样品均使胃癌细胞群体倍增时间延长 ,且使抑癌基因 p5 3蛋白表达增高 ;且AGPⅡ还使抑癌基因Fas及Fas L蛋白表达增高。 结论 该多糖抑制胃癌细胞的增殖 ,其抑癌机制与抑癌基因 p5 3、Fas及Fas L蛋白表达有关。
Objective To study the inhibitory effect of Acanthopanax senticosus polysaccharide on gastric cancer cells and its mechanism of action. Methods The doubling time of the cell population was measured to understand the effect of the polysaccharide on the proliferation of gastric cancer cells. The effect of the polysaccharide on the expression of the tumor suppressor gene p53, Fas and Fas L was detected by flow cytometry. Results The polysaccharides of Acanthopanax senticosus polysaccharides increased the doubling time of gastric cancer cell population and increased the expression of tumor suppressor gene p53 protein. AGPII also increased the expression of Fas and Fas L protein. Conclusion The polysaccharide inhibits the proliferation of gastric cancer cells and the mechanism of tumor suppressor is related to the expression of p53, Fas and Fas L proteins.
