目的 探索红毛五加多糖的体外抑癌机制。方法 采用体外细胞培养方法观察细胞形态学改变 ,采用流式细胞术检测细胞凋亡百分比及细胞周期改变。结果 红毛五加多糖使胃癌细胞出现凋亡的形态学改变。该多糖作用后在细胞周期直方图上出现凋亡峰 ,0 .1mmol/L红毛五加多糖样品Ⅱ (AGPⅡ )作用 36h、红毛五加多糖样品Ⅲ (AGPⅢ )作用 48h后 ,凋亡细胞分别占11.2 %和 15 .5 % ,并使细胞周期明显阻滞于G0 /G1期 ,具有时间—效应关系 ,且AGPⅡ的作用效果较AGPⅢ更为明显。结论 该多糖的抑癌机制与诱导细胞凋亡作用有关。
Objective To explore the anti-cancer mechanism of polysaccharides from Acanthopanax giraldii in vitro. Methods The cell morphology was observed by in vitro cell culture method. The percentage of cell apoptosis and cell cycle changes were detected by flow cytometry. Results The polysaccharides of Acanthopanax giraldii increased the morphological changes of apoptosis in gastric cancer cells. Apoptotic peaks appeared in the cell cycle histogram after the polysaccharides were treated. After a dose of 0.1 mmol/L AGP II for 36 h and AGP III for 48 h, apoptotic cells were observed. They accounted for 11.2% and 15.5%, respectively, and significantly blocked the cell cycle in the G0/G1 phase, with a time-effect relationship. The effect of AGPII was more pronounced than that of AGPIII. Conclusion The anti-cancer mechanism of this polysaccharide is related to the induction of apoptosis.
