目的:探讨表没食子儿茶素没食子酸酯(EGCG)对人胃癌MGC-803细胞异种接种的裸鼠肿瘤的抑制作用及其可能的机制。方法:对人胃癌MGC-803细胞异种接种成功的裸鼠腹腔注射不同剂量的EGCG,每天观察裸鼠的一般状态,处死后称重法测定肿瘤抑制率;HE染色观察肿瘤组织内细胞变化;RT-PCR测定Caspase-3 mRNA表达水平;免疫组化法测定Bcl-2、Bax表达;Western blot检测Smo、Gli-1蛋白表达。结果:与模型组相比EGCG(5、10、20mg/kg)对裸鼠肿瘤的抑制率分别为20.4%、21.2%和48.7%;与模型组相比,不同剂量的EGCG不同程度上调Caspase-3 mRNA表达和Bax表达,下调Bcl-2表达,抑制Smo、Gli-1蛋白表达。结论:EGCG可能通过Hedgehog信号抑制人胃癌MGC-803细胞接种的裸鼠移植瘤生长,诱导其凋亡。
AIM: To investigate the inhibitory effect of epigallocatechin-3-gallate (EGCG) on tumor inoculation of human gastric cancer cell line MGC-803 and its possible mechanism. Methods: Different doses of EGCG were intraperitoneally injected into nude mice successfully inoculated with human gastric cancer MGC-803 cells. The general state of nude mice was observed daily. The tumor inhibition rate was measured by weighing method after sacrificed. The changes of cell cycle were observed by HE staining. The expression of Caspase-3 mRNA was detected by PCR, the expression of Bcl-2 and Bax by immunohistochemical method, and the expression of Smo and Gli-1 by Western blot. Results: Compared with the model group, the inhibitory rates of EGCG (5, 10 and 20 mg / kg) on tumor in nude mice were 20.4%, 21.2% and 48.7%, respectively. Compared with the model group, EGCG at different doses upregulated Caspase- 3 mRNA expression and Bax expression, down-regulation of Bcl-2 expression, inhibition of Smo, Gli-1 protein expression. Conclusion: EGCG may inhibit the growth of xenografted human nude mice bearing human gastric cancer MGC-803 cells through Hedgehog signaling and induce its apoptosis.
