目的探讨水通道蛋白1(AQP1)和水通道蛋白8(AQP8)在维吾尔族妇女宫颈癌发生、浸袭过程中的表达及意义。方法选择2008年11月至2010年8月新疆医科大学附属肿瘤医院维吾尔族宫颈炎患者42例、宫颈上皮内瘤变(CIN)Ⅲ36例,宫颈癌98例。采用实时荧光定量RT-PCR方法和Western blot对其中10例宫颈炎和27例宫颈癌进行AQP1和AQP8 mRNA和蛋白检测;采用免疫组化对全部宫颈癌患者98例、CINⅢ36例及宫颈炎42例AQP1微血管密度和AQP8阳性率进行分析。结果 AQP1和AQP8在宫颈炎、宫颈癌早期(Ⅰb或Ⅱa期)及晚期(Ⅲ期)均有表达。宫颈炎组和宫颈癌早期、晚期组AQP1和AQP8 mRNA表达差异无统计学意义(P>0.05),而蛋白表达差异有统计学意义(P<0.01)。AQP1微血管密度和AQP8阳性表达率在宫颈炎、CINⅢ和宫颈癌组织中比较差异有统计学意义(P<0.01)。结论 AQP1和AQP8在宫颈癌发生、侵袭过程中可能发挥重要的生理作用。
Objective To investigate the expression and significance of aquaporin-1 (AQP1) and aquaporin-8 (AQP8) in Uighur women with cervical cancer. Methods From November 2008 to August 2010, 42 Uighur cervicitis patients, Cancer Center Hospital of Xinjiang Uygur Autonomous Region Cancer Hospital, 36 cases of cervical intraepithelial neoplasia (CIN) Ⅲ, 98 cases of cervical cancer. AQP1 and AQP8 mRNA and protein were detected in 10 cases of cervicitis and 27 cases of cervical cancer by real-time fluorescence quantitative RT-PCR and Western blot. Immunohistochemistry was used to detect 98 cases of cervical cancer, 36 cases of CINⅢ and 42 cases of cervicitis AQP1 microvessel density and AQP8 positive rates were analyzed. Results AQP1 and AQP8 were expressed in cervicitis, early cervical cancer (stage Ⅰb or Ⅱa) and in advanced stage (Ⅲ). There were no significant differences in the expression of AQP1 and AQP8 between cervicitis group and cervical cancer group at early stage and late stage (P> 0.05), but there was a significant difference in protein expression (P <0.01). AQP1 microvessel density and AQP8 positive expression rate in cervicitis, CIN Ⅲ and cervical cancer was significantly different (P <0.01). Conclusion AQP1 and AQP8 may play important physiological roles in the development and invasion of cervical cancer.
