通过比较细胞因子诱导的外周血来源的慢性乙型肝炎患者树突状细胞 (DC )和正常人DC在免疫分子表达和免疫功能等方面的差别 ,探讨慢性乙型肝炎患者DC所处的功能状态。以细胞计数、间接荧光表型分析、MTT法测定DC对同种异体淋巴细胞的刺激作用、ELISA法测定DC分泌IL 1 2、IL 6等方法进行研究。实验结果表明 :慢性乙型肝炎患者相同数量的前体细胞经诱导分化形成的DC数量明显减少 ;在培养的 3、 6、 9d,与同期正常人相比 ,慢性乙型肝炎患者的DC表达CD1、CD83、CD80和HLA DR分子水平均较低 ,而CD1 4分子表达水平较高 ;刺激同种异体淋巴细胞增殖能力也低于同期正常人 ;在培养的第 6天DC分泌的上清液中 ,慢性乙型肝炎患者DC分泌的IL 1 2水平低于正常人 ,而分泌的IL 6水平高于正常人。结果提示 ,慢性乙肝患者外周血来源的DC免疫功能处于抑制状态 ,细胞因子表达异常。
To compare the difference of immune molecule expression and immune function of dendritic cells (DCs) and normal human DCs from peripheral blood-derived chronic hepatitis B patients induced by cytokines, to explore the functional status of DCs in patients with chronic hepatitis B . The cytotoxicity of DCs to allogeneic lymphocytes was determined by MTT assay and the levels of IL-12 and IL-6 secreted by DCs were measured by cell counting and indirect fluorescent phenotype analysis. The experimental results showed that the number of DCs induced by the differentiation of the same number of precursor cells in patients with chronic hepatitis B was significantly decreased. Compared with normal subjects at 3, 6 and 9 days, DCs in chronic hepatitis B patients expressed CD1 , CD83, CD80 and HLA DR molecules were lower, while CD14 expression was higher. The ability of stimulating allogeneic lymphocyte proliferation was also lower than that of normal people in the same period. In the supernatant of DC secreted on the 6th day of culture , The level of IL-12 secreted by DCs in patients with chronic hepatitis B is lower than that of normal people, while the level of IL-6 secreted is higher than that of normal people. The results suggest that the immune function of DCs from peripheral blood of patients with chronic hepatitis B is in an inhibitory state and the expression of cytokines is abnormal.
