为了研究NKT细胞是否参与变应性鼻炎(allergic-rhinitis,AR)发病或通过细胞因子调控而影响该疾病发生,本研究通过分析AR患者外周血NKT细胞数量和增殖和产生细胞因子格局的动态变化来探索NKT在AR发病中的调节作用。通过用流式细胞仪分析其特异性NKT细胞表面标志发现,AR患者和正常人的外周血NKT细胞数量无明显差异。继之用NKT细胞特异性刺激剂α-GalCer进行NKT细胞增殖的动态分析,结果也提示AR患者和正常人的外周血NKT细胞增殖格局无明显差异。但是,用ELISA分析了AR患者外周血NKT细胞受α-GalCer刺激后分泌细胞因子格局后发现:AR患者的NKT细胞分泌IL-4和IL-5在72 h、96 h时比正常对照组明显增加,而分泌IL-12和IFN-γ的量则明显低于正常对照组。提示AR患者的NKT细胞可能通过分泌IL-4和IL-5上调Th2细胞功能,同时分泌IL-12和IFN-γ能力降低,提示AR患者的NKT细胞诱导Treg和自身的调节能力均降低。这些因素的综合可能是NKT细胞参与AR发病的重要途径之
In order to study whether NKT cells are involved in the pathogenesis of allergic rhinitis (AR) or affect the occurrence of the disease through the regulation of cytokines, this study analyzed the dynamic changes of NKT cell number, proliferation and cytokine pattern in peripheral blood of AR patients To explore the role of NKT in the pathogenesis of AR. However, the levels of IL-4 and IL-5 secreted by NKT cells in AR patients at 72 h and 96 h were significantly higher than those in normal controls Increased, while the amount of IL-12 and IFN-γ secretion was significantly lower than the normal control group. It is suggested that NKT cells in AR patients may upregulate the function of Th2 cells by secreting IL-4 and IL-5, while the ability of secreting IL-12 and IFN-γ is decreased.
