目的:研究miRNA-203在甲状腺乳头状癌组织中的表达水平及其与癌组织临床病理参数的相关性,并探讨其在甲状腺乳头状癌WRO细胞增殖过程中的作用。方法:收集2013-2016年30例甲状腺乳头状癌组织及其配对癌旁组织病理标本,采用荧光定量PCR方法检测甲状腺癌组织和癌旁组织中miRNA-203的表达水平,并分析其与临床病理参数的相关性;同时转染miRNA-203mimics调控WRO细胞株,用MTT、流式分析和Western blot方法分析WRO细胞的增殖能力、周期改变及相关周期蛋白(CyD1、CyB1)的变化。结果:相对于癌旁组织,甲状腺乳头状癌组织中miRNA-203显著低表达;在WRO细胞中转染miRNA-203mimics后,可显著抑制细胞增殖能力,阻滞G2/M期,同时上调CyB1、下调CyD1周期相关蛋白的表达。结论:甲状腺乳头状癌组织中miRNA-203可能起着抑癌作用,与甲状腺乳头状癌细胞的增殖能力相关,可能是甲状腺乳头状癌干预的新靶点之一。
OBJECTIVE: To study the expression of miRNA-203 in papillary thyroid carcinoma and its correlation with the clinicopathological parameters of papillary thyroid carcinoma and to explore the role of miRNA-203 in the proliferation of papillary thyroid carcinoma WRO cells. Methods: Thirty cases of papillary thyroid carcinoma and paired paracancerous tissues were collected from 2013 to 2016. The expression of miRNA-203 in thyroid carcinoma tissues and paracancerous tissues was detected by real-time fluorescence quantitative PCR, and its relationship with clinical pathology (WRO) cells were transfected with miRNA-203mimics. The proliferation and cell cycle changes of WRO cells and the changes of CyD1 and CyB1 were analyzed by MTT, flow cytometry and Western blot. Results: The miRNA-203 expression in papillary thyroid carcinoma was significantly lower than that in paracancerous tissues. After transfection with miRNA-203mimics in WRO cells, the miRNA-203mimics could significantly inhibit the cell proliferation, arrest the G2 / M phase and up-regulate the expression of CyB1, Down-regulate CyD1 cycle-related protein expression. Conclusions: miRNA-203 may play a role in tumor suppressor of papillary thyroid carcinoma, which is related to the proliferation of thyroid papillary carcinoma cells and may be one of the new targets of thyroid papillary carcinoma.
