目的探讨RNA结合蛋白人抗原R(HuR)和转录因子缺氧诱导因子-1α(HIF-1α)在肝癌组织中的表达及其与患者预后的关系。方法收集55例行肝癌手术切除后保存完好的肝癌组织和癌旁组织蜡块,同时选取20例正常人的肝脏组织作为正常对照,采用免疫组化染色法检测不同组织中HuR和HIF-1α蛋白表达阳性率,采用Spearman相关分析研究肝癌组织中HuR和HIF-1α蛋白表达的相关关系,采用Kaplan-Meier法生存分析肝癌组织HuR和HIF-1α蛋白表达与患者预后的关系。结果肝癌组织、癌旁组织和正常肝组织中HuR蛋白的阳性表达率分别为67.27%、20.0%和5.0%,3种组织中HuR蛋白阳性表达率的差异具有统计学意义(P<0.01);肝癌组织、癌旁组织和正常肝组织中HIF-1α蛋白的阳性表达率分别为58.18%、12.73%和10.0%,肝癌组织中HIF-1α蛋白阳性表达率高于癌旁组织和正常肝组织,差异有统计学意义(P<0.01)。经Spearman相关分析,肝癌组织HuR蛋白表达与HIF-1α蛋白表达呈正相关(rs=0.508,P<0.01);Log-rank检验表明,HuR蛋白表达阳性的肝癌患者的5年随访中位无瘤生存期为27个月,显著低于HuR蛋白表达阴性患者的32个月(log-rank=19.350,P<0.01);HIF-1α蛋白表达阳性的肝癌患者的5年随访中位无瘤生存期为28个月,显著低于HIF-1α蛋白表达阴性患者的37个月(log-rank=26.556,P<0.01)。结论肝癌组织中HuR和HIF-1α表达率异常增高,两者在肝癌组织中的表达呈正相关,且与患者的无瘤生存时间密切相关。
Objective To investigate the expression of human binding protein R (HuR) and transcription factor hypoxia inducible factor-1α (HIF-1α) in human hepatocellular carcinoma (HCC) and its relationship with prognosis. Methods 55 cases of hepatocellular carcinoma after resection of the well-preserved liver cancer tissue and adjacent paraffin blocks were collected at the same time, 20 cases of normal human liver tissue as a normal control, immunohistochemical staining of HuR and HIF-1α protein in different tissues The positive rates of HuR and HIF-1α in hepatocellular carcinoma were analyzed by Spearman correlation analysis. The relationship between the expression of HuR and HIF-1α in hepatocellular carcinoma and prognosis was analyzed by Kaplan-Meier method. Results The positive rates of HuR protein in HCC tissue, paracancer tissue and normal liver tissue were 67.27%, 20.0% and 5.0%, respectively. There was significant difference in the expression of HuR protein between the three tissues (P <0.01). The positive rate of HIF-1α protein expression in HCC tissue, paracancerous tissue and normal liver tissue were 58.18%, 12.73% and 10.0% respectively. The positive rate of HIF-1α protein in HCC tissue was higher than that in paracancerous tissue and normal liver tissue, The difference was statistically significant (P <0.01). Spearman correlation analysis showed that there was a positive correlation between HuR protein expression and HIF-1α protein expression (rs = 0.508, P <0.01). Log-rank test showed that patients with HuR positive hepatocellular carcinoma had a median follow-up of 5 years (P <0.01). The 5-year follow-up median tumor-free survival of patients with HIF-1α protein expression was significantly lower than that of patients with negative HuR protein expression for 32 months (log-rank = 19.350, 28 months, significantly lower than those in patients with negative HIF-1α protein expression for 37 months (log-rank = 26.556, P <0.01). Conclusions The expression of HuR and HIF-1α in HCC tissues is abnormally increased. The expression of HuR and HIF-1α in HCC tissues is positively correlated with the expression of HuR and HIF-1α.
