目的:探讨组织蛋白酶B(CB)特异性siRNA对人食管癌EC9706细胞体外侵袭力的抑制作用。方法:将体外转录合成的针对CB的特异性siRNA设10μmol/L、15μmol/L和20μmol/L3个不同剂量转染人食管癌EC9706细胞,用Boydenchamber体外侵袭实验检测转染前后细胞体外侵袭力的变化。对照组设无关siRNA转染组、空白对照组(转染空脂质体)及正常对照组(不进行任何操作)。结果:和正常对照组相比,特异性siRNA15μmol/L组和20μmol/L组穿越Matrigel胶的细胞数明显减少(P<0.05),而10μmol/L特异性siRNA组、空白对照组和无关siRNA转染组与正常对照组之间差异均无统计学意义(P>0.05)。结论:特异性CBsiRNA能有效抑制人食管癌EC9706细胞的体外侵袭力。
Objective: To investigate the inhibitory effect of cathepsin B (CB) specific siRNA on in vitro invasiveness of human esophageal carcinoma EC9706 cells. Methods: Specific siRNA targeting CB was transfected into human EC9706 cells with different doses of 10μmol / L, 15μmol / L and 20μmol / L. The in vitro invasiveness of the transfected cells was detected by Boydenchamber invasion assay Variety. In the control group, no siRNA transfection group, blank control group (transfected with empty liposome) and normal control group (without any operation). Results: Compared with the normal control group, the number of cells in 15μmol / L siRNA group and 20μmol / L group significantly decreased (P <0.05), while 10μmol / L specific siRNA group, blank control group and unrelated siRNA transfection There was no significant difference between the dye group and the normal control group (P> 0.05). Conclusion: Specific CBsiRNA can effectively inhibit the invasiveness of EC9706 cells in vitro.
