目的:复制大鼠移植性肝癌模型,探讨扶正抗癌方对大鼠肝癌的疗效及机制。方法:采用Walker256癌肉瘤接种于SD大鼠肝脏,制作移植肝癌模型,分为正常组、假手术组、模型组、扶正抗癌方低、中、高剂量组。记录体质量等生命体征;实验1、2、3w分别检测血清TB、DB、UB;称取瘤重、测量瘤块长短径;取瘤组织、瘤旁组织行病理观察;记录肿瘤转移情况。结果:扶正抗癌方各剂量组大鼠生命体征好于模型组,术后10d模型组大鼠体质量增长加速,可能与腹水产生有关,术后18d高剂量组体质量低于模型组(P<0.05);各剂量中药组胆红素明显低于模型组(P<0.05或P<0.01);扶正抗癌方可抑制肿瘤的生长,其中高剂量组肿瘤质量抑制率高于低剂量组(P<0.05);病理结果示扶正抗癌方可促进癌细胞坏死,抑制其向周围浸润;各剂量组转移评分低于模型组(P<0.05)。结论:扶正抗癌方具有改善移植性肝癌大鼠生存状态、降低胆红素、促进癌细胞坏死、抑制肿瘤生长转移的作用,其中高剂量组效果最突出。
OBJECTIVE: To replicate rat transplanted hepatocellular carcinoma (HCC) models and explore the efficacy and mechanism of Fuzheng Kangai Recipe on rat liver cancer. Methods: The liver of SD rats was inoculated with Walker256 carcinosarcoma and the transplanted hepatocellular carcinoma model was established. The model was divided into normal group, sham operation group, model group, Fuzheng Kangnafang low-, middle-, and high-dose groups. The vital signs such as body mass were recorded; serum TB, DB, and UB were measured at 1, 2 and 3 w, respectively; the tumor weight was weighed, and the length and length of the tumor block were measured; tumor tissue and paratumor tissue were taken for pathological observation; and the tumor metastasis was recorded. Results: The vital signs of rats in Fuzheng Kangaifang group were better than those in the model group. The body weight of rats in the model group increased at 10 days after operation, which may be related to the occurrence of ascites. The body weight of the high-dose group at the 18th day after operation was lower than that of the model group. <0.05); The dose of bilirubin in the traditional Chinese medicine group was significantly lower than that in the model group (P<0.05 or P<0.01); Fuzheng Kangai recipe could inhibit the growth of the tumor, and the high-dose group had higher tumor suppression than the low-dose group ( P <0.05); pathological results showed that Fuzheng Kangai Fang can promote cancer cell necrosis and inhibit its infiltration into the surrounding; the dose of each dose group was lower than the model group (P <0.05). Conclusion: Fuzheng Kangaifang can improve the survival state of transplanted hepatocellular carcinoma rats, reduce bilirubin, promote cancer cell necrosis, inhibit tumor growth and metastasis, among which the high dose group has the most prominent effect.
