目的 :探讨黄芩素(baicalein,BAI)对胃癌MGC-803细胞凋亡的影响。方法 :不同浓度BAI处理MGC-803细胞后,应用MTT法检测细胞的增殖情况,FCM法检测细胞的凋亡,Hoechst 33342染色观察细胞中凋亡小体的形成,蛋白质印迹法检测细胞中凋亡相关蛋白的表达。结果:5、10、15、25和50μmol/L BAI可明显抑制胃癌MGC-803细胞的增殖(P值均<0.05);10、15和25μmol/L BAI处理组MGC-803细胞凋亡率明显升高(P值均<0.05);Hoechst 33342染色后可见10、15和25μmol/L BAI处理组MGC-803细胞中出现的凋亡小体数量增加(P值均<0.05);BAI可显著上调MGC-803细胞中剪切型caspase-3和剪切型聚腺苷二磷酸-核糖聚合酶(cleaved-poly ADP-ribose polymerase,cleaved-PARP)蛋白的表达水平(P值均<0.01),下调MGC-803细胞中Bcl-2蛋白表达水平(P<0.01)。结论:BAI可有效抑制胃癌MGC-803细胞的增殖,并诱导细胞凋亡,其机制可能与BAI调节caspase-3、PARP和Bcl-2等凋亡相关蛋白的表达有关。
Objective: To investigate the effect of baicalein (BAI) on the apoptosis of gastric cancer cell line MGC-803. Methods: MGC-803 cells were treated with different concentrations of BAI, the cell proliferation was detected by MTT assay, the apoptosis of cells was detected by FCM, the formation of apoptotic bodies was observed by Hoechst 33342 staining and the apoptosis was detected by Western blotting Related protein expression. Results: 5, 10, 15, 25 and 50μmol / L BAI could significantly inhibit the proliferation of gastric cancer MGC-803 cells (all P <0.05). The apoptotic rates of MGC-803 cells treated with 10, 15 and 25μmol / (P <0.05). After Hoechst 33342 staining, the numbers of apoptotic bodies in MGC-803 cells treated with BAI at 10, 15 and 25μmol / L increased (P <0.05), BAI increased significantly The expressions of cleaved-poly (ADP-ribose) polymerase (cleaved-PARP) and the protein of cleaved-poly (ADP-ribose polymerase) in MGC-803 cells were significantly decreased The expression of Bcl-2 protein in MGC-803 cells (P <0.01). CONCLUSION: BAI can effectively inhibit the proliferation and induce apoptosis of gastric cancer cell line MGC-803, which may be related to the regulation of BAI on the expression of apoptosis related proteins such as caspase-3, PARP and Bcl-2.
