目的研究十二指肠和小肠良恶性肿瘤组织中碱性成纤维细胞生长因子(bFGF)信使核糖核酸(mRNA)、血小板源生长因子(PDGF)mRNA和血小板吸附蛋白1(TSP-1)mRNA的表达与微血管计数(MVC)的关系及其临床病理意义。方法MVC染色应用SABC免疫组织化学法,bFGFmRNA、PDGFmRNA和TSP-1mRNA染色为原位杂交染色法。结果腺癌MVC(46.2±7.4)明显高于腺瘤(19.8±8.1)。腺癌bFGFmRNA和PDGFmRNA阳性率(57%,48%)明显高于腺瘤(20%,30%),而TSP-1mRNA则相反(腺癌36%,腺瘤90%)。MVC、bFGFmRNAPDGFmRNA表达阳性高与肿瘤分化差、浸润深、直径大及淋巴结转移有关。腺癌中bFGFmRNA表达与PDGFmRNA表达呈正相关,但与TSP-1mRNA表达呈负相关。结论bFGF、PDGF和TSP-1均为重要的肿瘤血管生成调节因子,其mRNA表达水平与十二指肠和小肠腺癌发生发展、生物学行为及转移的发生有密切关系。
Objective To study the expressions of bFGF mRNA, PDGF mRNA and TSP-1 mRNA in duodenum and small bowel benign and malignant tumors. Expression and microvascular count (MVC) and its clinicopathological significance. Methods MVC staining SABC immunohistochemistry, bFGFmRNA, PDGFmRNA and TSP-1mRNA staining for in situ hybridization staining. Results The adenocarcinoma MVC (46.2 ± 7.4) was significantly higher than adenoma (19.8 ± 8.1). The positive rates of bFGF mRNA and PDGF mRNA in adenocarcinoma (57%, 48%) were significantly higher than those in adenomas (20%, 30%), while those in TSP-1 mRNA were opposite (adenocarcinoma 36%, adenoma 90%). The positive expression of MVC and bFGFmRNAPDGFmRNA correlated with poor differentiation, deep infiltration, large diameter and lymph node metastasis. The expression of bFGF mRNA in adenocarcinoma was positively correlated with the expression of PDGF mRNA, but negatively correlated with the expression of TSP-1 mRNA. Conclusions Both bFGF, PDGF and TSP-1 are important tumor angiogenesis regulators. The mRNA expression levels of bFGF, PDGF and TSP-1 are closely related to the development, biological behavior and metastasis of duodenal and intestinal adenocarcinoma.
