目的 :通过大鼠胸主动脉腹腔移植模型 ,探讨移植物动脉硬化 (CAA)形成过程中增生内、外膜平滑肌细胞IGF 1R浕 表达情况 ,并观察表达程度与平滑肌细胞增殖程度的关系。方法 :雄性健康纯系Wistar和SD大鼠分别作为供体和受体 ,分为急性排斥 (A组 )、免疫耐受组和同系对照组。取腹腔移植胸主动脉标本进行Masson和免疫组化染色 ,比较移植术后不同时间内、外膜VSMC增生和纤维化程度以及IGF 1R浕 在增生外膜和内膜平滑肌细胞上的表达水平 ,IGF 1R浕 与浕 actin和PCNA表达程度的关系。结果 :CAA发生后 ,浕 actin阳性细胞出现在外膜 ,外膜增生和纤维化程度与IGF 1R浕 和PCNA的表达程度一致 ,增生内膜表面有浕 actin、IGF 1R浕 和PCNA表达。术后A组外膜IGF 1R浕 的表达程度逐渐增高 (P <0 .0 1)。术后第 8周IGF 1R浕 的表达程度与浕 actin和PCNA相当。结论 :IGF 1R浕 在CAA平滑肌细胞中高度表达 ,表达程度与血管尤其外膜平滑肌细胞增生程度密切相关。
OBJECTIVE: To investigate the expression of IGF-1R in proliferative and adventitial smooth muscle cells (VSMCs) during the development of arteriosclerosis (CAA) in rats through rat thoracic aorta peritoneal transplantation model and to observe the relationship between the expression level and the proliferation of smooth muscle cells. Methods: Male healthy healthy Wistar and SD rats were used as donors and recipients respectively and divided into acute rejection group (A group), immunocompromised group and homologous control group. The specimens of abdominal aorta were collected for Masson and immunohistochemical staining. The expression of VSMC proliferation and fibrosis in vitro and the expression of IGF1R in proliferating adventitia and endometrial smooth muscle cells were compared at different time points after transplantation. IGF 1R 浕 and 浕 actin and PCNA expression levels. RESULTS: After CAA, 浕 actin positive cells appeared on the adventitia, and the degree of adventitial hyperplasia and fibrosis was consistent with the expression of IGF1R 浕 and PCNA. The expression of 浕 actin, IGF1R 浕 and PCNA on proliferative endometrium was observed. Postoperative A group of outer membrane IGF 1R 逐 expression gradually increased (P <0.01). The expression of IGF-1R at the 8th week was similar to that of actin and PCNA. CONCLUSION: IGF1R 高 is highly expressed in CAA smooth muscle cells and its expression is closely related to the degree of vascular smooth muscle cell proliferation, especially.
