目的探讨半胱氨酰白三烯受体1(Cys LTsR1)拮抗剂孟鲁司特对肠缺血再灌注(I/R)损伤的保护作用。方法大鼠肠I/R模型,大鼠随机分为假手术组、模型组、孟鲁司特(2 mg·kg-1体重)组,检测各组小肠含水量变化,末端脱氧核苷酸转移酶介导的d UTP缺口末端标记测定法(TUNEL)检测各组小肠黏膜细胞的凋亡,蛋白质印迹法检测各组Cys LTsR1、半胱氨酸天冬氨酸蛋白酶9(caspase 9)和半胱氨酸天冬氨酸蛋白酶8(caspase 8)的表达的变化。结果与模型组相比,孟鲁司特组小肠含水量、肠组织黏膜细胞凋亡、Cys LTR1、caspase 9和caspase 8的表达水平均明显降低(P<0.05)。结论孟鲁司特可以减少肠I/R肠黏膜细胞的凋亡,抑制caspase 8和caspase 9的表达可能是重要的凋亡机制。
Objective To investigate the protective effect of montelukast on human intestinal ischemia reperfusion (I / R) injury induced by Cys LTsR1 antagonist. Methods Intestinal I / R model rats were randomly divided into sham-operation group, model group and montelukast (2 mg · kg-1 body weight) group. Conclusion Montelukast can reduce the intestinal I / R intestinal mucosa apoptosis, inhibiting caspase 8 and caspase 9 expression may be an important mechanism of apoptosis.
