【目的】观察丹参酮ⅡA 对胃癌细胞增殖的影响及其与环氧合酶-2(COX-2)表达的关系。【方法】体外培养胃癌细胞株 SGC7901,经丹参酮ⅡA 作用后,采用四唑盐(MTT)比色法检测细胞增殖,流式细胞仪检测细胞凋亡,免疫细胞化学 SABC 法检测 COX-2蛋白表达,放射免疫法检测前列腺素 E2(PGE2)含量。【结果】丹参酮ⅡA 对胃癌细胞的生长有明显的抑制作用,并呈剂量依赖性。以0.5 mg/L 作用终浓度抑制作用最明显,其48 h 的抑制率为71.1%,与对照组相比差异有显著性(P <0.01);5 mg/L 丹参酮ⅡA 作用后,随时间的延长,凋亡率逐渐升高,48 h 达到高峰,随后逐渐下降,与对照组比较,各处理组都有显著性差异(P <0.05);丹参酮作用组胃癌细胞 COX-2表达明显减少,其培养液中 PGE2的产生量下降,与对照组相比差异均有显著性(P <0.01)。【结论】丹参酮ⅡA 可能是通过下调 COX-2 mRNA 的表达水平发挥其对胃癌细胞生长抑制及促进凋亡作用。“,”[Objective]To observe the effect of TanshinoneⅡA on proliferation and apoptosis of human gastric carci-noma cell line SGC7901 and its correlation with COX-2 expression.[Methods]Human gastric carcinoma cell line SGC7901 was cultured in vitro and treated with Tanshinone ⅡA .The cell proliferation was detected by MTT assay,and the cell ap-optosis was detected by flow cytometry,the expression of COX-2 protein was detected by immunohistochemistry SABC method,and the content of prostaglandin E2 (PGE2)was detected by radioimmunoassay.[Results]The growth of gastric cancer cells was significantly inhibited by TanshinoneⅡA ,and dose dependence there occurred.The inhibition effect of 0.5 mg/L was the most obvious dosage,and the inhibition rate of 48 h reached 71.1%,the difference was significant com-pared with the control group (P <0.01);With 5 mg/L of TanshinoneⅡA trestment ,the apoptosis rate increased grad-ually with the time and reached its peak at 48th h,then gradually decreased.Compared with the control group,the treat-ment group had significant difference (P <0.05);COX-2 expression in gastric cancer cells of the treatment group was significantly decreased,and the production of PGE2 in the culture fluid decreased significantly (P <0.01).[Conclusion]TanshinoneⅡA may play a role in the growth inhibition and apoptosis of gastric cancer cells by down regulating the expres-sion of COX-2 mRNA.
