目的:研究海带中岩藻聚糖硫酸酯(Fucoidan,Fu)的抗氧化活性及对非酒精性脂肪性肝病(Nonalcoholic fatty liver disease,NAFLD)的保护作用。方法:取雄性清洁级SD大鼠60只,随机均分为6组,分别为正常对照组、高血脂模型组、阳性对照组(辛伐他汀1.0 mg/kg)、Fu低剂量组(25 mg/kg)、中剂量组(50 mg/kg)和高剂量组(100 mg/kg)。正常对照组饲喂标准饲料,其他组饲喂高脂饲料(即在普通饲料的基础上加100 g/L猪油和20 g/L胆固醇)。实验为期30 d。实验结束后麻醉各组大鼠,留取血清,测定血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL);取肝组织,测定匀浆上清液超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)含量,并做病理组织学检查。结果:50 mg/kg和100 mg/kg Fu可显著减轻高血脂模型动物血生化指标(P<0.05,P<0.01),改善肝组织中SOD、MDA和CAT氧化指标(P<0.05,P<0.01)。此外,与模型组比较,50 mg/kg和100 mg/kg Fu可明显减轻高脂所致大鼠肝细胞脂肪变性、减小肝组织及胞浆中脂肪空泡和细胞体积。结论:Fu对大鼠NAFLD具有一定的保护作用,其作用机制可能与其抗氧化活性有关。
Objective: To study the antioxidant activity of fucoidan (Fu) in kelp and its protective effect on nonalcoholic fatty liver disease (NAFLD). Methods: Sixty male SD rats were randomly divided into 6 groups: normal control group, hyperlipidemia model group, positive control group (simvastatin 1.0 mg / kg), Fu low dose group (25 mg / kg), middle dose (50 mg / kg) and high dose (100 mg / kg). The normal control group was fed the standard diet, while the other groups were fed the high fat diet (ie, 100 g / L lard and 20 g / L cholesterol on the basis of normal diet). Experimental period of 30 d. At the end of the experiment, rats in each group were anesthetized, serum was taken, serum triglyceride (TG), total cholesterol (TC), high density lipoprotein (HDL) and low density lipoprotein (LDL) were measured; Serum superoxide dismutase (SOD), malondialdehyde (MDA), catalase (CAT) content, and histopathological examination. Results: 50 mg / kg and 100 mg / kg Fu could significantly reduce the blood biochemical indexes (P <0.05, P <0.01) and improve the indexes of SOD, MDA and CAT oxidation in hyperlipidemia model rats (P < 0.01). In addition, compared with the model group, 50 mg / kg and 100 mg / kg Fu could significantly reduce the fatty degeneration of rat hepatocytes induced by high fat and reduce the lipid vacuoles and cell volume in the liver and cytoplasm. Conclusion: Fu has certain protective effect on NAFLD in rats, and its mechanism may be related to its antioxidant activity.
