目的:探讨黑素瘤抗原(melanoma-associated antigen,MAGE)-A4及肿瘤抑制基因P73在乳腺癌组织和相应癌旁组织中的表达及其临床意义。方法:选取60例河北医科大学第四医院2007年9月至2007年12月乳腺癌住院患者,采用免疫组织化学法检测乳腺癌组织及相应癌旁组织中MAGE-A4和P73蛋白的表达,并分析其与乳腺癌临床病理特征的相关性。结果:乳腺癌组织中MAGE-A4、P73蛋白阳性表达率为63.3%(38/60)、43.3%(26/60),癌旁组织中MAGE-A4、P73蛋白阳性表达率为0(0/60)、85%(51/60)(均P<0.01)。乳腺癌组织中MAGE-A4蛋白的表达与P73蛋白的表达呈正相关(r=0.316,P=0.014)。MAGE-A4蛋白的表达与乳腺癌患者的年龄、病理类型、组织学分级、临床分期、肿瘤大小、淋巴转移、雌激素受体(estrogen receptor,ER)表达、孕激素受体(progestrogen receptor,PR)表达均无相关性,但与C-erbB2蛋白的表达呈负相关(r=-0.259,P=0.046)。P73蛋白表达与乳腺癌患者的年龄、病理类型、组织学分级、临床分期、肿瘤大小、淋巴结转移、CerbB2表达均无相关性,但与ER(r=0.274,P=0.034)和PR的表达呈正相关(r=0.262,P=0.043)。结论:乳腺癌组织高表达MAGE-A4和P73蛋白,且MAGE-A4与P73蛋白之间可能存在重要的相互调节机制。
Objective: To investigate the expression and clinical significance of melanoma-associated antigen (MAGE) -A4 and tumor suppressor gene P73 in breast cancer tissues and corresponding paracancerous tissues. Methods: Sixty-six inpatients with breast cancer from September 2007 to December 2007 in the Fourth Hospital of Hebei Medical University were selected for immunohistochemical detection of MAGE-A4 and P73 protein in breast cancer tissues and corresponding paracancerous tissues Analysis of its correlation with clinicopathological features of breast cancer. Results: The positive rates of MAGE-A4 and P73 in breast cancer tissues were 63.3% (38/60) and 43.3% (26/60), respectively. The expression of MAGE-A4 protein was correlated with age, pathological type, histological grade, clinical stage, tumor size, lymph node metastasis, estrogen receptor (ER) expression, progestrogen receptor ), But negatively correlated with the expression of C-erbB2 protein (r = -0.259, P = 0.046). There was no correlation between the expression of P73 protein and the age, pathological type, histological grade, clinical stage, tumor size, lymph node metastasis and CerbB2 expression in patients with breast cancer, but it was positively correlated with ER (r = 0.274, P = 0.034) (R = 0.262, P = 0.043). Conclusion: MAGE-A4 and P73 proteins are highly expressed in breast cancer tissues, and MAGE-A4 and P73 proteins may have important inter-regulatory mechanisms.
