目的 研究白介素-1(IL-1)预刺激对反义受体相关激酶-2寡核苷酸(IRAK-2 ODN)抑制IL-1激活核因子-kappa B(NF-κB)的影响。方法 实验分IL-1预处理组和非预处理组,以脂质体介导反义 IRAK-2 ODN转染293细胞,以夹心酶联免疫吸附测定法(ELISA)检测NF-κB的含量。结果 IL-1非预处理组反义IRAK-2 ODN不能抑制NF-(B活化,而预处理组NF-κB活化受到明显抑制(4 (g 反义 IRAK-2 ODN与细胞共孵育8 h时,NF-κB的光密度值从ODN转染前的0.835±0.013下降到转染后的 0.261±0.008),且这种抑制作用具有时间(5~24 h)和剂量(1~8 (g)依赖性。结论 IL-1预刺激在反义IRAK-2 ODN抑制IL-1诱导的NF-κB活化中起决定性作用。
Objective To investigate the effect of interleukin-1 (IL-1) preconditioning on the inhibition of IL-1-activated nuclear factor-kappa B (NF-κB) by antisense receptor-associated kinase-2 oligonucleotide (IRAK-2 ODN). Methods The IL-1 preconditioning and non-preconditioning groups were transfected into 293 cells with liposome-mediated antisense IRAK-2 ODN. The levels of NF-κB were detected by sandwich enzyme linked immunosorbent assay (ELISA). Results The antisense IRAK-2 ODN of non-pretreatment with IL-1 could not inhibit the activation of NF- (B, but the activation of NF-κB in pretreatment group was significantly inhibited , The optical density of NF-κB decreased from 0.835 ± 0.013 before ODN transfection to 0.261 ± 0.008 after transfection), and the inhibitory effect had time (5 ~ 24 h) and dose (1 ~ 8 (g) Dependent manner.Conclusion IL-1 preconditioning plays a decisive role in the inhibition of IL-1-induced NF-κB activation by antisense IRAK-2 ODN.
