目的建立啤酒及果汁中8种生物胺(组胺、酪胺、腐胺、尸胺、精胺、亚精胺、色胺、2-苯乙胺)的高效液相色谱法(HPLC)。方法采用丹磺酰氯衍生,以1,7-二氨基庚烷为内标,C18柱(250 mm×4.6 mm ID,5μm)分离,二极管阵列检测器波长254 nm检测。结果本法8种生物胺标准曲线线性相关系数均大于0.999,方法检出限为0.2 mg/kg(组胺为0.4 mg/kg),平均回收率为72.57%~104.7%,RSD为2.51%~5.12%。结论本法操作简便、定量准确可靠。
OBJECTIVE To establish a high performance liquid chromatography (HPLC) method for the determination of eight biogenic amines (histamine, tyramine, putrescine, cadaverine, spermine, spermidine, tryptamine, 2- phenylethylamine) in beer and fruit juices. Methods Dansyl chloride derivatized with 1,7-diaminoheptane as an internal standard was separated on a C18 column (250 mm × 4.6 mm ID, 5 μm) and detected by a diode array detector at 254 nm. Results The linear correlation coefficients of eight biogenic amines calibration curves were all greater than 0.999. The detection limits were 0.2 mg / kg (histamine 0.4 mg / kg), the average recovery was 72.57% -104.7% and the RSD was 2.51% 5.12%. Conclusion This method is simple, accurate and reliable.
