目的:探讨重肌灵片(ZJL)对细胞因子水平的影响。方法:将50只Lewis大鼠随机等量分为佐剂对照组、模型组、地塞米松组及重肌灵大、小剂量组,用从电鳐电器官提取的烟碱型乙酰胆碱受体(N2AchR)纯品,加完全弗氏佐剂(CFA)免疫大鼠制成实验性自身免疫性重症肌无力(EAMG)。大鼠免疫后第24天,用ELISA检测细胞培养上清及血清中IFN-γ、IL-10和TGF-β的含量。结果:在细胞培养上清中,模型组与CFA组比较IFN-γ、IL-10及TGF-β的水平升高(P<0.01);ZJL两剂量组与模型组比较IFN-γ、IL-10的含量均显著下降(P<0.01或P<0.05);而TGF-β的含量均显著升高(P<0.01或P<0.05)。阳性药组与模型组比较,三者含量均显著降低(P<0.01或P<0.05)。结论:ZJL片可通过上调TGF-β的含量,抑制IFN-γ、IL-10的过量分泌,从而降低抗N2AchR抗体的水平,抑制EAMG的发生。
Objective: To investigate the effect of ZJL on the level of cytokines. METHODS: Fifty Lewis rats were randomly and equally divided into adjuvant control group, model group, dexamethasone group, and large and small doses of digoxin group. The nicotinic acetylcholine receptors extracted from the electric organs of the rats were used. Pure N2AChR) rats were immunized with complete Freund’s adjuvant (CFA) to produce experimental autoimmune myasthenia gravis (EAMG). On the 24th day after immunization of rats, the cell culture supernatant and serum IFN-γ, IL-10 and TGF-β levels were detected by ELISA. RESULTS: In the cell culture supernatant, the levels of IFN-γ, IL-10, and TGF-β were increased in the model group and the CFA group (P<0.01); IFN-γ and IL- were compared between the two groups of the ZJL group and the model group. The content of 10 significantly decreased (P<0.01 or P<0.05), while the content of TGF-β increased significantly (P<0.01 or P<0.05). Compared with the model group, the content of the three drugs in the positive drug group was significantly decreased (P<0.01 or P<0.05). Conclusion: ZJL tablets could inhibit the excessive secretion of IFN-γ and IL-10 by up-regulating the content of TGF-β, thus reducing the level of anti-N2AchR antibody and inhibiting the occurrence of EAMG.
