目的 探讨重肌灵片对实验性自身免疫性重症肌无力 (EAMG)体内一氧化氮 (NO)和一氧化氮合成酶 (NOS)的影响。方法 用从电鳐电器官提取的含N2 AchR粗提液加完全弗氏佐剂免疫Lewis大鼠制成EAMG ,ELISA法检测大鼠免疫后第 7周血清及细胞培养上清中IFN γ含量。采用比色法检测血清、淋巴细胞培养液和肌肉组织中的NO和NOS。结果 模型组与完全弗氏佐剂 (CFA)组比较干扰素 γ(IFN γ)、NO和NOS水平升高 (P <0 0 1 ) ,重肌灵组与模型组比较IFN γ、NO和NOS含量下降 (P <0 0 1 )。结论 重肌灵下调EAMG体内IFN γ和AChRab含量导致NO和NOS合成减少 ,从而降低由NO引起的肌肉神经组织损伤
Objective To investigate the effects of rencrelin tablets on nitric oxide (NO) and nitric oxide synthase (NOS) in experimental autoimmune myasthenia gravis (EAMG). Methods EAMG was prepared by immunizing Lewis rats with crude extracts containing N2 AchR extracted from electrocautery organs plus complete Freund’s adjuvant. IFN-γ levels in serum and cell culture supernatants at 7 weeks after immunization were measured by ELISA. Colorimetry was used to detect NO and NOS in serum, lymphocyte culture fluid, and muscle tissue. Results Compared with the complete Freund’s adjuvant (CFA) group, the levels of IFN γ, NO, and NOS were increased in the model group (P < 0 01). The IFN γ, NO, and NOS in the graoskin group were compared with the model group. The content decreased (P < 0 01). Conclusion The down-regulation of IFN-γ and AChRab in EAMG by Shengjiling results in a decrease in the synthesis of NO and NOS, which reduces the damage of nerve tissue caused by NO.
