应用卵蛋白致敏和刺激小鼠复制过敏性哮喘模型观察血栓素A2合成酶选择性抑制剂OKY-046对抗原引起呼吸道嗜酸性粒细胞(EOS)浸润的影响。结果表明正常对照小鼠支气管肺泡灌洗液(BALF)中未见到EOS;致敏小鼠给予抗原反复刺激后,BALF中EOS急剧增多。OKY-046的不同剂量分别导致BALF中EOS数下降36.9%,58.0%及68.9%。还发现OKY-046抑制EOS对气道的侵润伴随着IL-5和IL-2水平的明显降低,这些结果提示OKY-046通过抑制CD4T淋巴细胞分泌细胞因子从而抑制了EOS在气道的积聚。
The effect of OKY-046, a selective inhibitor of thromboxane A2 synthase, on the eosinophilic respiratory tract eosinophil (EOS) infiltration was observed using the ovalbumin-sensitized and stimulated mouse model of allergic asthma. The results showed that EOS was not found in BALF in normal control mice. EOS in BALF increased sharply after sensitized mice repeatedly received antigens. Different doses of OKY-046 resulted in a decrease of 36.9%, 58.0% and 68.9% in BALF. It was also found that OKY-046 inhibited EOS infiltration in the airway accompanied by a significant decrease of IL-5 and IL-2 levels. These results suggest that OKY-046 inhibits the accumulation of EOS in the airway by inhibiting the secretion of cytokines by CD4 T lymphocytes .
