目的探讨大鼠脑梗死后内质网应激与少突胶质细胞相关因子髓鞘碱性蛋白(myelin basic protein,MBP)的关系。方法 60只雄性SD大鼠随机分为3组,每组20只,脑梗死组、4-苯基丁酸(4-PBA)组采用手术线栓法制备大鼠大脑中动脉梗死模型,对照组不行大脑中动脉结扎;造模成功4.5、6、24、72h,各组分别取5只大鼠脑组织采用免疫组织化学方法检测脑组织葡萄糖调节蛋白78(glucose regulated protein 78,GRP78)、MBP,比较不同时间点各组GRP78、MBP阳性细胞率。结果脑梗死组4.5、6、24h时GRP78阳性细胞率[(65.00±8.89)%、(74.00±7.00)%、(71.33±15.56)%]高于对照组[(33.00±7.21)%、(35.33±5.03)%、(32.33±4.72)%](P<0.05),72h时GRP78阳性细胞率[(50.67±12.01)%]与对照组[(38.67±2.08)%]比较差异无统计学意义(P>0.05);脑梗死组4.5、6、24、72h时GRP78阳性细胞率与4-PBA组[(65.00±7.81)%、(78.33±7.03)%、(86.33±3.78)%、(56.67±13.01)%]比较差异无统计学意义(P>0.05);脑梗死组4.5、6、24、72h时MBP阳性细胞率[(47.33±6.42)%、(42.67±3.51)%、(32.67±6.65)%、(30.00±1.00)%]均低于对照组[(61.33±4.72)%、(61.33±9.86)%、(58.33±9.71)%、(58.33±10.40)%](P<0.05);4-PBA组4.5h时MBP阳性细胞率[(46.00±8.18)%]低于脑梗死组,6、24、72h时MBP阳性细胞率[(48.33±8.50)%、(45.67±13.79)%、(37.67±2.08)%]高于脑梗死组,但差异均无统计学意义(P>0.05)。结论 MBP在大鼠脑梗死后4.5、6、24、72h的表达呈下降趋势,且不被内质网应激抑制剂4-PBA所抑制。
Objective To investigate the relationship between endoplasmic reticulum stress and oligodendrocyte-related factor myelin basic protein (MBP) after cerebral infarction in rats. Methods Sixty male Sprague-Dawley rats were randomly divided into three groups (20 rats in each group). The cerebral infarction model was established by surgical thread occlusion in cerebral infarction group and 4-phenylbutyric acid (4-PBA) group. The control group No middle cerebral artery ligation was made; the success of model establishment was 4.5,6,24,72h, the brain tissues of 5 rats in each group were taken to detect the expression of glucose regulated protein 78 (GRP78), MBP, The percentage of GRP78 and MBP positive cells in each group was compared at different time points. Results Compared with control group [(33.00 ± 7.21)%, (35.33%), (65.00 ± 8.89)%, (74.00 ± 7.00)%, (71.33 ± 15.56)%, (50.67 ± 12.01)%] compared with that of the control group [(38.67 ± 2.08)%]] (P <0.05). There was no significant difference in the percentage of GRP78 positive cells at 72h (P <0.05). Compared with 4-PBA group, the percentage of GRP78-positive cells in the infarcted group was significantly higher than that in the 4-PBA group at 4.5,6,24,72h [(65.00 ± 7.81)%, (78.33 ± 7.03)%, (86.33 ± 3.78)%, (P> 0.05). The percentage of MBP positive cells in the infarction group at the time points of 4.5, 6, 24 and 72 h [(47.33 ± 6.42)%, (42.67 ± 3.51)% and (32.67 ± 6.65 ) Were significantly lower than those in the control group [(61.33 ± 4.72)%, (61.33 ± 9.86)%, (58.33 ± 9.71)%, (58.33 ± 10.40)%, respectively] The rate of MBP-positive cells in the 4-PBA group was (46.00 ± 8.18)% at 4.5 h, lower than that in the cerebral infarction group (48.33 ± 8.50)%, (45.67 ± 13.79)% at 6, (37.67 ± 2.08)%] higher than those in cerebral infarction group, but the difference was not statistically significant (P> 0.05). Conclusion The expression of MBP at 4.5, 6, 24 and 72 h after cerebral infarction in rats showed a decreasing trend and was not inhibited by endoplasmic reticulum stress inhibitor 4-PBA.
