目的:研究川芎总黄酮(LCTF)的提取和抗氧化作用。方法:总黄酮的提取采用微波提取和索氏提取2种方法,设置不同的因素水平,从而确定最佳提取方案;抗氧化研究采用总黄酮对血清抗活性氧能力的影响,对小鼠肝匀浆生成MDA的影响,对脂肪氧合酶活性的影响,总黄酮对H2O2诱导小鼠氧化溶血的影响。结果:微波提取总黄酮最佳因素提取频率为530W,提取时间为20min,70%乙醇,液固比为15∶1,得率为1.008%。抗氧化作用表明,2.00g.L-1LCTF对脂肪氧合酶的粗酶活性抑制率超过50%;LCTF在0.5~10.0g.L-1范围内,具有较强的体外抗活性氧能力,并能减少小鼠肝匀浆MDA的生成,可抑制H2O2诱导的小鼠红细胞溶血。结论:微波提取LCTF得率较高,且LCTF具有较强的抗氧化能力,并与总黄酮溶液的浓度具有剂量-效应关系。
Objective: To study the extraction and antioxidation of total flavonoids from ligusticum chuanxiong (LCTF). Methods: The extraction of total flavonoids by microwave extraction and Soxhlet extraction two kinds of methods, set different levels of factors to determine the best extraction program; antioxidant studies using flavonoids on serum anti-reactive oxygen species, liver homogenate Pulp formation MDA, the impact of lipoxygenase activity, the impact of total flavonoids on H2O2-induced oxidative hemolysis in mice. Results: The optimal extraction frequency of total flavonoids from microwave was 530W, extraction time was 20min, 70% ethanol, liquid-solid ratio was 15:1, yield was 1.008%. Antioxidant activity showed that 2.00gL-1LCTF inhibited the lipoxygenase crude enzyme activity by more than 50%; LCTF was in the range of 0.5-10.0gL-1, had strong ability of resisting reactive oxygen species in vitro, The formation of liver homogenate MDA can inhibit the hemolysis of mouse erythrocytes induced by H2O2. CONCLUSION: The yield of LCTF extracted by microwave is high, and LCTF has a strong antioxidant capacity and a dose-response relationship with the concentration of total flavonoids.
