目的研究蛋白酶激活受体-1(PAR-1)在骨肉瘤中的表达情况及其与微血管密度(MVD)的相关性,探讨其在骨肉瘤侵袭转移中的作用。方法采用免疫组化法检测38例骨肉瘤及其癌旁组织中PAR-1的表达情况,以CD34为标记,计算MVD。结果 PAR-1阳性表达率及MVD在骨肉瘤组织中分别为89.4%、(42.5±12.8)条,显著高于癌旁组织中的55.3%、(14.8±6.5)条,且二者跟骨肉瘤的Enneking分期、软组织浸润、肺转移有关(P均<0.05)。PAR-1与MVD呈正相关(r=0.748,P<0.01)。结论 PAR-1在骨肉瘤肿瘤组织中存在过度表达,且与肿瘤微血管密度呈正相关,与肿瘤侵袭转移、血管生成相关。
Objective To investigate the expression of PAR-1 in osteosarcoma and its relationship with microvessel density (MVD), and to explore the role of PAR-1 in the invasion and metastasis of osteosarcoma. Methods Immunohistochemistry was used to detect the expression of PAR-1 in 38 cases of osteosarcoma and its adjacent tissues. CD34 was used to calculate MVD. Results The positive rates of PAR-1 and MVD in osteosarcoma tissues were 89.4% and 42.5 ± 12.8, respectively, which were significantly higher than those in adjacent tissues (55.3% and 14.8 ± 6.5) Enneking staging, infiltration of soft tissue and lung metastasis (all P <0.05). There was a positive correlation between PAR-1 and MVD (r = 0.748, P <0.01). Conclusions PAR-1 is overexpressed in osteosarcoma tissues and positively correlates with tumor microvessel density, which is correlated with tumor invasion and metastasis and angiogenesis.
