目的:探讨色素框同源物2(chromobox homolog 2,CBX2)在结直肠癌组织和细胞中的表达情况及临床意义。方法:采用实时荧光定量PCR和蛋白质印迹法检测结直肠癌细胞、正常结直肠上皮细胞、结直肠癌组织和癌旁组织中CBX2 mRNA和蛋白的表达水平;采用免疫组织化学法检测66例结直肠癌及其对应的癌旁正常组织中CBX2的表达情况。分析CBX2表达与结直肠癌患者临床病理特征及预后的关系。结果:结直肠癌细胞中CBX2 mRNA和蛋白的表达水平均高于正常结肠上皮细胞FHC(P值均<0.05)。结直肠癌组织中CBX2 mRNA和蛋白的表达水平均高于癌旁组织(P值均<0.05);CBX2在结直肠癌组织中的阳性表达率为53.0%(35/66),明显高于癌旁正常组织中的7.6%(5/66),差异有统计学意义(P<0.05)。CBX2表达与结直肠癌患者的TNM分期、淋巴结转移、远处转移及生存时间显著相关(P值均<0.05)。CBX2高表达的结直肠癌患者的总生存时间短于低表达患者(P=0.01)。CBX2表达和远处转移是结直肠癌患者预后的独立影响因素(P值均<0.05)。结论:结直肠癌组织和细胞中CBX2过表达,在一定程度上参与了结直肠癌的发生和发展。并且CBX2的表达是结直肠癌患者预后的独立影响因素。
Objective: To investigate the expression and clinical significance of chromobox homolog 2 (CBX2) in colorectal cancer tissues and cells. Methods: The expression of CBX2 mRNA and protein in colorectal cancer cells, normal colorectal epithelial cells, colorectal cancer tissues and adjacent normal tissues were detected by real-time fluorescence quantitative PCR and Western blotting. 66 cases of colorectal cancer were detected by immunohistochemistry Cancer and its corresponding adjacent normal tissue CBX2 expression. To analyze the relationship between CBX2 expression and clinicopathological features and prognosis in patients with colorectal cancer. Results: The expression of CBX2 mRNA and protein in colorectal cancer cells was higher than that of normal colorectal epithelial cells (P <0.05). The expression of CBX2 mRNA and protein in colorectal cancer was higher than that in adjacent non-cancerous tissues (P <0.05). The positive rate of CBX2 in colorectal cancer was 53.0% (35/66) 7.6% (5/66) in normal tissues, the difference was statistically significant (P <0.05). CBX2 expression was significantly associated with TNM stage, lymph node metastasis, distant metastasis and survival time in patients with colorectal cancer (all P <0.05). Patients with CBX2 overexpressing colorectal cancer had a shorter overall survival than patients with low expression (P = 0.01). CBX2 expression and distant metastasis are independent prognostic factors in patients with colorectal cancer (P all <0.05). Conclusion: CBX2 overexpression in colorectal cancer tissues and cells is involved in the occurrence and development of colorectal cancer to a certain extent. And CBX2 expression is an independent prognostic factor in patients with colorectal cancer.
