目的检测良性前列腺增生(BPH)患者组织中蛋白表达的特点,了解有无与BPH相关的特异性蛋白表达。方法通过双向电泳方法所发现的检测BPH组织特异性蛋白表达,αsl-Casein进行定量免疫组化研究来验证该蛋白表达的特异性。结果通过对正常前列腺组织,BPH及前列腺癌组织进行免疫组织化学染色,发现αsl-Casein在90%(20/22)的BPH组织中呈弱到强表达,而在正常前列腺组织中未见表达(0/10),在前列腺癌组织中的表达也不足10%(3/30)。结论αsl-Casein可能是一种BPH诊断的生物标记物。
Objective To detect the expression of protein in the tissues of patients with benign prostatic hyperplasia (BPH) and to find out whether BPH-related protein is expressed or not. Methods The expression of BPH tissue-specific protein was detected by two-dimensional electrophoresis. The specificity of αsl-Casein was examined by quantitative immunohistochemistry. Results Immunohistochemical staining of normal prostate tissue, BPH and prostate cancer showed that αsl-Casein was weakly and strongly expressed in BPH tissues of 90% (20/22), but not in normal prostate tissues ( 0/10) and less than 10% (3/30) in prostate cancer tissue. Conclusion αsl-Casein may be a biomarker for the diagnosis of BPH.
