目的:研究盐酸千金藤碱(CH)对鼻咽癌细胞系SUNE-1的生长抑制和诱导凋亡作用。方法:采用MTT法检测CH对SUNE-1细胞生长的抑制作用,分别用流式细胞分析仪、透射电镜观察CH处理SUNE-1细胞后细胞周期阻滞和细胞凋亡的发生。结果:MTT法证实,CH对SUNE-1细胞生长有抑制作用,且呈剂量-效应关系和时间-效应关系,P<0.01。流式细胞仪检测发现,CH处理SUNE-1细胞后,实验组G0/G1细胞(71.5±8.7)%明显高于对照组(46.9±10.5)%,P<0.05,提示细胞周期停滞于G0/G1期,流式细胞周期分布图可见凋亡峰。透射电镜可见,SUNE-1细胞出现凋亡细胞形态学改变。结论:CH对SUNE-1细胞有抑制作用,机制可能与阻滞细胞周期及诱导细胞凋亡相关。
Objective: To study the growth inhibition and induction of apoptosis of nasopharyngeal carcinoma cell line SUNE-1 by cepharanthine hydrochloride (CH). Methods: The inhibitory effect of CH on the growth of SUNE-1 cells was detected by MTT assay. Cell cycle arrest and apoptosis were observed by flow cytometry and transmission electron microscopy. Results: The results of MTT assay showed that CH could inhibit the growth of SUNE-1 cells in a dose-dependent and time-dependent manner (P <0.01). Flow cytometry analysis showed that the percentage of G0 / G1 cells in experimental group (71.5 ± 8.7%) was significantly higher than that in control group (46.9 ± 10.5%), P <0.05, suggesting that cell cycle arrest in G0 / G1 phase, apoptotic peak can be seen by flow cytometry. TEM showed that morphological changes of apoptotic cells appeared in SUNE-1 cells. CONCLUSION: CH inhibits SUNE-1 cells, which may be related to arresting cell cycle and inducing apoptosis.
