目的探讨大黄素对大鼠肾间质纤维化模型中血小板源性生长因子(PDGF-B)表达的影响及意义。方法清洁级Wister雄性成年大鼠160只随机分为4组:正常对照组、假手术组、模型组及大黄素治疗组。制备单侧输尿管梗阻(UUO)模型[1];治疗组在UUO的基础上每天以大黄素40mg/kg灌胃,分别在术后1d、3d、7d、14d,每组取10只大鼠处死,取左侧肾脏行免疫组织化学方法检测肾组织中PDGF-B的表达水平,动态观察PDGF-B在各组大鼠术后梗阻侧肾脏1、3、7、14天的组织学变化及大黄素对其影响。结果模型组PDGF-B的表达持续升高;治疗组PDGF-B的表达水平低于模型组(P<0.01)。结论中药大黄素可以通过减少PDGF-B的表达来发挥其延缓肾间质纤维化进程的作用,对肾纤维化有治疗作用。
Objective To investigate the effect of emodin on the expression of platelet-derived growth factor (PDGF-B) in rats with renal interstitial fibrosis and its significance. Methods Totally 160 Wister male rats were randomly divided into 4 groups: normal control group, sham operation group, model group and emodin treatment group. Unilateral ureteral obstruction (UUO) model was established [1]. The expression of PDGF-B in the treatment group was lower than that in the model group (P <0.01). Conclusion Emodin can play a role in delaying the process of renal interstitial fibrosis by reducing the expression of PDGF-B and has a therapeutic effect on renal fibrosis.
