目的 研究探索三氧化二砷(0.5μ mol/L作用6小时)对恶性淋巴细胞基因表达的影响。方法 采用细胞培养,mRNA的提取和分离,制备荧光探针和基因芯片技术。结果 在基因芯片的200个凋亡基因中,有10%(20/200)的基因出现差异性表达,其中18个凋亡基因下调,2个基因出现上调。值得注意的是,凋亡抑制蛋白,同性连锁的凋亡抑制蛋白下调。P53和其他的凋亡基因也下调。在上调的基因中,热休克蛋白有促进Raji细胞凋亡的作用。结论 凋亡抑制蛋白在Raji细胞的凋亡中,起重要的作用。
Objective To explore the effect of arsenic trioxide (0.5 μmol / L for 6 hours) on the gene expression of malignant lymphocytes. Methods Cell culture, mRNA extraction and isolation were used to prepare fluorescent probes and gene chip technology. Results Among the 200 apoptotic genes in the gene chip, 10% (20/200) genes were differentially expressed, of which 18 were down-regulated and two were up-regulated. It is noteworthy that, apoptosis inhibitor protein, same-sex chain inhibitor of apoptosis protein down. P53 and other apoptotic genes are also down-regulated. In the upregulated genes, heat shock proteins promote the apoptosis of Raji cells. Conclusion The apoptosis-inhibiting protein plays an important role in the apoptosis of Raji cells.
