目的探讨健脾化瘀方抑制人胃癌MGC-803细胞侵袭转移的作用机制。方法分别采用Real-time q PCR和Western Blot技术检测健脾化瘀方对胃癌MGC-803细胞侵袭转移基因转录和蛋白表达的影响。结果健脾化瘀方作用于人胃癌MGC-803细胞24h后,MMPs、VEGF随着给药浓度增加其mRNA和蛋白表达下降,RECK随着给药浓度增加其mRNA和蛋白表达上升,STAT3基因转录和蛋白表达未见明显变化,而p-STAT3蛋白表达量随着给药浓度增加下降。结论健脾化瘀方具有体外抑制MGC-803细胞侵袭转移的作用,其分子机制可能与抑制STAT3蛋白磷酸化和RECK基因转录、表达,进而影响下游VEGF、MMPs等有关。
Objective To investigate the mechanism of Jianpi Huayu Recipe inhibiting invasion and metastasis of human gastric cancer cell line MGC-803. Methods Real-time q PCR and Western Blot were used to detect the effect of Jianpi Huayu Recipe on the gene transcription and protein expression of invasion and metastasis in gastric cancer cell line MGC-803. Results After treated with Jianpi Huayu Recipe for 24h, the mRNA and protein expressions of MMPs and VEGF decreased with the increase of drug concentration, and the expression of RECK mRNA and protein increased with the increase of drug concentration. The transcription of STAT3 gene And no significant changes in protein expression, while p-STAT3 protein expression decreased with increasing concentration. Conclusion Jianpi Huayu Recipe can inhibit the invasion and metastasis of MGC-803 cells in vitro. The molecular mechanism may be related to the inhibition of STAT3 phosphorylation and the transcription and expression of RECK gene, and further influence of downstream VEGF and MMPs.
