目的:比较中药联合骨髓间充质干细胞BMSC或骨髓单个核细胞BMNC蛛网膜下腔移植对缺血再灌脑损伤脑组织中血管内皮生长因子含量DE的影响。方法:将分离纯化的BMSCS和BMNCS以蛛网膜下腔注射的方法移植到缺血再灌脑损伤大鼠颅内,同时进行中药联合治疗。用定量ELISA的方法检测移植后4天2、8天脑匀浆内VEGF含量。结果:BMNC组和BMSC组在4天时VEGF含量变化无统计学意义,在28天时VEGF含量显著升高,且BMNC组升高更显著;中药联合BMNC组和中药联合BMSC组在4天、28天时VEGF含量均显著升高,且比单纯的BMNC组和BMSC组VEGF含量显著升高,中药联合BMNC组比中药联合BMSC组VEGF含量显著升高。治疗早期和后期比较,中药联合BMSC组含量降低无统计学意义。结论:BMSC和BMNC经蛛网膜下腔移植可在治疗28天左右明显提高MCAO大鼠脑组织中VEGF含量,中药联合BMNC或BMSC治疗可使VEGF水平升高更提前、幅度更高、效果更平稳而持久。中药与BMNC联合在提高MCAO大鼠脑组织中VEGF含量方面更显优势。
Objective: To compare the effects of BMSC with traditional Chinese medicine (BMSC) or bone marrow mononuclear cells (BMNC) on the changes of vascular endothelial growth factor (DE) in brain tissue after ischemia / reperfusion injury. Methods: The isolated and purified BMSCS and BMNCS were subcutaneously injected into the intracranial cerebral ischemia reperfusion injury in rats, while Chinese medicine combined therapy. The content of VEGF in brain homogenate was detected by ELISA on days 2 and 8 after transplantation. Results: There was no significant difference in VEGF content between BMNC group and BMSC group at 4 days. VEGF content increased significantly at 28th day in BMNC group and BMNC group, while BMNC group and BMSC group increased significantly at 4 days and 28 days VEGF levels were significantly increased, and than simple BMNC group and BMSC group VEGF content was significantly increased, Chinese medicine combined with BMNC group than the traditional Chinese medicine combined with BMSC group VEGF increased significantly. Compared with the BMSC group, there was no significant difference between the early treatment and the later treatment. CONCLUSIONS: BMSC and BMNC can significantly increase the content of VEGF in the brain tissue of MCAO rats after 28 days of treatment, and the combination of BMNC and BMSC can increase the level of VEGF more quickly and with a more stable effect And lasting. Chinese medicine and BMNC combined to improve the VEGF content in brain tissue of MCAO rats.
