本文对鱼类人工雌核发育中两个技术处理进行了优化分析;对精子的照射时间、照射条件和卵子染色体加倍的温度休克等技术进行了最佳选择。目的在于推广应用于生产。在30w×2支紫外线灯、照射距离10cm、精液厚度0.2mm条件下,最佳照射时间为900秒左右;热休克和冷休克诱导鱼卵染色体加倍均可获得一定数量的雌核二倍体鱼;在生产上进行大量鱼卵处理时,鲤鱼采用4℃低温休克1,000秒、草鱼4℃低温休克900秒是可行的。
In this paper, two techniques of artificial gynogenetic development in fish were optimized, and the optimal selection of sperm irradiation time, irradiation conditions and temperature shock doubling of egg chromosomes were carried out. The purpose is to promote the use of production. Under the conditions of 30w × 2 ultraviolet lamps, irradiation distance of 10 cm and semen thickness of 0.2 mm, the optimal irradiation time was about 900 seconds. A number of female diploid fish could be obtained by doubling the chromosomes induced by heat shock and cold shock ; In the production of a large number of fish eggs treatment, carp cryogenic shock at 4 ℃ for 1,000 seconds, grass carp 4 ℃ low temperature shock of 900 seconds is feasible.
