调控miR-3686的表达对胰腺癌细胞系生物学行为的影响

  目的 探讨调控miR-3686的表达对胰腺癌细胞系生物学行为的影响.方法 qRT-PCR和Western-blot检测胰腺癌细胞系PANC1和永生化胰腺上皮细胞HPDE6C7中miR-3686和PLK1的表达水平.人工合成miR-3686mimicS和miR-3686scramble,选用处于对数生长期PANC1细胞,按照分组分别进行转染MTT实验和克隆形成实验检测各组PANC1细胞增殖的变化.移植瘤实验检测各组裸鼠移植瘤生长的情况.结果 胰腺癌细胞系中,miR-3686在PANC1细胞低表达,但在HPDE6C7细胞中高表达(P<0.01),而PLK1结果相反.移植瘤实验结果显示,miR-3686组裸鼠移植瘤的生长在转染后2w后明显受到抑制.处死后剥离肿瘤,结果显示miR-3686组肿瘤体积显著小于scramble组和Mock组肿瘤体积(P<0.01).结论 体外上调miR-3686的表达能抑制胰腺癌细胞系PANC1细胞的生长以及侵袭,可促进胰腺癌细胞的凋亡,并影响胰腺癌细胞的周期变化.裸鼠体内上调miR-3686的表达能够抑制移植瘤生长.
