  In apple peel,there are two synthesis pathways of UDP-galactose.One is UDP-glucose pathway catalyzed by UDP-glucose 4-epimerase(UGE),the other is galactose pathway catalyzed by galactose-1-phosphate uridine transferase.However,metabonomics analysis showed that galactose was undetectable in apple peel.Therefore,UGE may be responsible for biosynthesis of UDP-galactose.It indicated that UGE could be in favour of anthocyanin synthesis and accumulation for its higher activity in red apple peel than that of the non-red peel.In this study,MdUGE2 was isolated from fruit peel of blush mutant ofFujiapple(Malus×domestica Borkh.).The full-length of MdUGE2 was 1335 bp.It had an open reading frame(ORF)of 1038 bp,encoding 345 amino acid residues.The phylogenetic tree analysis revealed that amino acids sequence of MdUGE2 shared 97.0%similarity with predicted UGE GEPI48-like from pear cultivarDangshansuli(Pyrus x bretschneideri)but 66.9%similarity with MdUGE1 from fruit peel of apple cultivarTsugaru(Malus domestica).Real-time quantitative PCR analysis showed that the expression level of MdUGE2 in stems,leaves,flowers,peels and fleshes of mutant blushedFujiwas higher than that of the stripedFuji.The most relative expression quantity of MdUGE2 from peel and pulp of mutant blushedFujiwere 64.1%and 21.7%higher than that of stripedFujirespectively.At the same time,the relative expression quantity of MdUGE2 appeared two peaks on 152 DAFB and 156 DAFB in peel of mutant blushedFujiand striped Fuji,on the first peak,the relative expression quantity of MdUGE2 from peel mutant blushedFujiwas about twice as large as that of stripedFuji,while on the second peak,the former was about five times as the latter.The results indicated that the expression level of MdUGE2 was closely related with anthocyanin accumulation and distribution of apple peel.
