目的从细胞凋亡因子Fas、Fas-L、Bax、Bcl-2角度探讨安胃汤对慢性萎缩性胃炎模型大鼠疗效的作用机制。方法采用甲基硝基亚硝基胍(MNNG)慢性萎缩性胃炎大鼠模型,应用安胃汤进行干预;运用HE染色观察胃黏膜组织形态改变;ELISA方法检测血清中Fas-L含量;采用实时荧光定量PCR方法、Western blot检测Fas、Bax、Bcl-2基因表达及其蛋白含量的水平。结果该方可以明显上调Fas、Bax基因以及其蛋白含量的表达,下调外周血Fas-L的含量和Bcl-2基因以及其蛋白含量的表达。结论安胃汤能够明显改善CAG大鼠胃组织的病理变化,具有明显的抗CAG作用,其作用机制可能通过提高Fas基因及蛋白的表达,降低Fas-L含量,激活CAG大鼠胃黏膜细胞Fas/Fas-L信号传导通路,降低Bcl-2基因及蛋白,升高Bax基因及蛋白,促进胃黏膜细胞凋亡,可能是安胃汤防治CAG的关键机制之一。
Objective To explore the mechanism of AnWei Decoction on curative effect of chronic atrophic gastritis model rats from the perspective of apoptosis factors Fas, Fas-L, Bax and Bcl-2. Methods A rat model of Chronic Atrophic Gastritis with Methylnitroso-nitrosoguanidine (MNNG) was used. An An-tang decoction was used for intervention. The morphology of gastric mucosa was observed by HE staining. The content of Fas-L in serum was detected by ELISA. The levels of Fas, Bax, Bcl-2 gene and their protein levels were detected by real-time fluorescence quantitative PCR and Western blot. Results The formula could significantly upregulate the expression of Fas and Bax gene and its protein content, and downregulate the expression of Fas-L and Bcl-2 gene and protein in peripheral blood. Conclusion Anwei Decoction can significantly improve the pathological changes of gastric tissue in CAG rats and has a significant anti-CAG effect. Its mechanism may be through increasing the expression of Fas gene and protein, decreasing the content of Fas-L, activating Fas / Fas-L signaling pathway, decrease Bcl-2 gene and protein, increase Bax gene and protein and promote gastric mucosal cell apoptosis, which may be one of the key mechanisms of AnWei Decoction in preventing and treating CAG.
