目的研究大孔吸附树脂分离纯化鹰嘴豆芽总异黄酮的工艺参数。方法以鹰嘴豆芽总异黄酮为考察指标,对8种大孔树脂采用静态吸附方法,优选出吸附解吸性能最佳的大孔树脂,并优化纯化条件,确定最佳工艺参数。结果 HPD-300型树脂对鹰嘴豆芽总异黄酮有较好的吸附和解吸效果。其最佳纯化条件为:上样质量浓度3.05 mg/m L,上样体积4.5 BV,上样体积流量为2 BV/h;6 BV蒸馏水除杂后,继用5 BV20%乙醇洗脱除杂;洗脱剂95%乙醇,洗脱用量6 BV,洗脱体积流量2 BV/h。按此工艺条件验证试验,总异黄酮质量分数达到41.1%。结论 HPD-300型大孔树脂适合富集纯化鹰嘴豆芽总异黄酮。
Objective To study the process parameters of separation and purification of total isoflavones from chickpea sprouts by macroporous adsorption resin. Methods Taking the total isoflavones of garbanzo bean sprouts as the index, the macroporous resin with the best adsorption and desorption properties was selected for the eight kinds of macroporous resins by static adsorption method. The optimal purification conditions were determined and the optimum technological parameters were determined. Results HPD-300 resin had good adsorption and desorption effects on the total isoflavones of chickpea sprouts. The optimal purification conditions were as follows: loading mass concentration of 3.05 mg / m L, loading volume of 4.5 BV, loading volume of 2 BV / h; 6 BV distilled water, then 5 BV20% ; Eluent 95% ethanol, elution 6 BV, elution volume flow 2 BV / h. According to this process conditions validation test, the total isoflavone content of 41.1%. Conclusion HPD-300 macroporous resin is suitable for enrichment of total flavonoids of purified chickpea sprouts.
