目的:研究何首乌不同炮制品对大鼠肝细胞色素P4502E1(CYP2E1)基因mRNA表达的影响。方法:50只大鼠随机分为空白组、清蒸组、黑豆汁制首乌A、B、C 5组,给药组每日灌胃主药5g.kg-1,空白组连续给同等体积生理盐水,连续给药90d,实验结束时采用逆转录聚合酶链反应(RT-PCR)法测定大鼠肝CYP2E1基因mRNA在给药后的变化。结果:各给药组CYP2E1基因mRNA表达水平明显低于空白组(P<0.01),各给药组组间差异无显著性(P>0.05)。结论:首乌不同炮制品均能够显著抑制CYP2E1mRNA的表达。
Objective: To study the effect of different processed products of Polygonum multiflorum on mRNA expression of hepatocyte cytochrome P4502E1 (CYP2E1) in rats. Methods: Fifty rats were randomly divided into blank control group, steamed group and black bean juice group A, B and C 5. The rats in the treatment group were fed with 5 g.kg-1 of the main drug daily and the blank group were given the same volume of physiology Saline, continuous administration 90d, the end of the experiment using reverse transcription polymerase chain reaction (RT-PCR) assay rat liver CYP2E1 mRNA expression changes after administration. Results: The mRNA expression of CYP2E1 in each group was significantly lower than that in the blank group (P <0.01). There was no significant difference between the two groups (P> 0.05). Conclusion: Polygonum multiflorum processed products can significantly inhibit the expression of CYP2E1 mRNA.
