目的观察姜黄素(curcumin)对人胶质瘤细胞U87增殖的抑制作用,及其对细胞中TGF-β1/Smad4蛋白表达的影响,探讨curcumin抗胶质瘤的分子机制。方法体外培养U87细胞,用不同浓度的curcumin(5、10、20、40μmol/L)作用不同时间(24、48和72 h)后,采用MTT法检测U87细胞生长情况,流式细胞术检测细胞周期的变化,Western blot法检测细胞中转化生长因子β1(transforming growth factorβ1,TGF-β1)及其下游因子Smad4蛋白的表达水平。结果低剂量curcumin对U87细胞生长无明显的抑制作用,细胞周期也无明显变化;高剂量curcumin能显著降低U87细胞的存活率(P<0.05),明显降低S期细胞比例,增加G0期细胞比例,出现了明显的G0/G1期阻滞,而TGF-β1和Smad4蛋白的水平也明显受到高浓度curcumin的抑制,且这种抑制作用均呈浓度-时间依赖性(P<0.05)。结论 curcumin对体外生长的U87细胞的生长具有明显的抑制作用,其机制可能与抑制异常激活的TGF-β1/Smad4通路有关。本研究为临床治疗胶质瘤提供了新的思路。
Objective To investigate the inhibitory effect of curcumin on the proliferation of human glioma U87 cells and its effect on the expression of TGF-β1 / Smad4 in human glioma cells and to explore the molecular mechanism of curcumin against glioma. Methods U87 cells were cultured in vitro. The growth of U87 cells was detected by MTT assay after different concentrations of curcumin (5, 10, 20, 40 micromol / L) for 24, 48 and 72 h. Flow cytometry Western blot was used to detect the expression of transforming growth factor β1 (TGF-β1) and its downstream Smad4 protein. Results Low dose curcumin had no obvious inhibitory effect on the growth of U87 cells and no significant changes of cell cycle. High dose curcumin could significantly reduce the survival rate of U87 cells (P <0.05), significantly reduce the proportion of S phase cells and increase the proportion of cells in G0 phase , And obvious G0 / G1 phase arrest occurred. The levels of TGF-β1 and Smad4 were also significantly inhibited by high concentration of curcumin, and the inhibitory effect was in a time-and concentration-dependent manner (P <0.05). Conclusion Curcumin has a significant inhibitory effect on the growth of U87 cells in vitro, which may be related to the inhibition of abnormal TGF-β1 / Smad4 pathway. This study provides a new idea for the clinical treatment of glioma.
