以人牙胚为材料,通过RNA原位杂交技术检测WNT/β-CATENIN信号通路的关键因子(受体LRP6,转导因子APC和DVL2)在人牙胚的帽状期和钟状期的表达模式.此外,利用Real-time PCR技术验证这些基因在钟状期的门牙和磨牙,以及磨牙的上皮和间充质的mRNA表达水平.结果显示,LRP6、APC和DVL2基因在帽状期和钟状期人牙胚的上皮和间充质均有表达,而且APC和DVL2基因在钟状期的门牙和磨牙的表达存在差异,LRP6和APC在牙胚上皮和间充质的表达存在差异.结果提示,WNT/β-CATENIN信号通路关键因子LRP6、APC和DVL2在人牙齿发育过程中存在重要的调控作用.
