通过构建人转化生长因子β1(hTGF-β1)的正、反义基因表达载体并通过基因转染技术,首次将hTGF-β1的正、反义基因分别导入到HL-60细胞中,经G418选择获得可稳定表达hTGF-β1的HL-60细胞。较为详细地研究了hTGF-β1的正、反义基因对HL-60细胞周期的影响及对细胞增殖的调控作用。结果表明,表达正义hTGF-β1 mRNA的HL-60细胞阻滞于G1期,而表达反义hTGF-β1 mRNA的HL-60细胞可以解除G1期的阻滞,使大部分细胞进入S期。加入正义hTGF-β1基因后,HL-60细胞形成集落的能力下降,细胞生长速度下降,恶变程度降低;加入反义hTGF-β1基因后,其结果正相反。
By constructing the positive and antisense gene expression vectors of human transforming growth factor-β1 (hTGF-β1) and by gene transfection technology, the positive and antisense genes of hTGF-β1 were introduced into HL-60 cells for the first time, and were selected by G418. HL-60 cells stably expressing hTGF-β1 were obtained. The effects of the positive and antisense genes of hTGF-β1 on the cell cycle of HL-60 cells and the regulation of cell proliferation were studied in detail. The results showed that HL-60 cells expressing sense hTGF-β1 mRNA were arrested at G1 phase, while HL-60 cells expressing antisense hTGF-β1 mRNA could be released from G1 phase arrest and most cells entered S phase. After adding the hTGF-β1 gene, the ability of HL-60 cells to form colonies decreased, the cell growth rate decreased, and the degree of malignant transformation decreased. When antisense hTGF-β1 gene was added, the result was the opposite.
