用高效液相色谱法测定人血浆中甲硫哒嗪的浓度。以氯氮平为内标,标本在碱性条件下用叔丁基甲醚(叔丁基甲醚与异丙醇之比为60:1,v/v)提取,流动相为甲醇:水(75:25,内含四甲基乙二胺、冰醋酸分别为0.45%,0.35%,v/v),用紫外检测器在254nm进行测定。线性范围为100~1000ng/ml,最低检出限为5ng/ml,绝对回收率为88.7±4.1%(±SD)天内与天间变异分别为4.6%和4.1%。本法简单、快速、灵敏、准确,适用于甲硫哒嗪的血药浓度监测及临床药理学研究。
Determination of Thioridazine in Human Plasma by High Performance Liquid Chromatography. Clozapine was used as an internal standard.
