目的:研究二脱甲氧基姜黄素(Bdmc)与阿霉素(ADR)联合应用对人白血病多药耐药细胞株(HL-60/ADR)的生长抑制作用。方法:HL-60/ADR细胞2×103/孔,1,2,4,8μmol·L-1Bdmc分别与0.4,0.8,1.6,3.2μmol·L-1ADR同时联合给药,48 h后,MTT法测定两种药物的体外杀伤作用。6,12,24,48μmol·L-1Bdmc分别与0.08,0.2,0.4,1.0μmol·L-1ADR序贯联合给药,48 h后,应用金氏公式进行序贯联合用药效果分析。结果:单独ADR 0.4,0.8μmol·L-1时抑制率6.9%,11.8%,同时联合加8μmol·L-1Bdmc时,抑制率达到94%以上。1~8μmol·L-1Bdmc和0.8~3.2μmol·L-1ADR同时联合作用HL-60/ADR细胞可产生单纯相加至增强的协同杀伤效果。序贯给药法中,先给Bdmc后给SDR结果为协同作用,先给ADR后给Bdmc实验结果为拮抗作用。结论:Bdmc与ADR同时用药可产生协同作用,二者序贯联合用药仅产生单向协同作用。
Objective: To study the inhibitory effect of didemethoxy curcumin (Bdmc) combined with adriamycin (ADR) on the growth of multidrug-resistant human leukemia cell line HL-60 / ADR. METHODS: HL-60 / ADR cells were treated with 2 × 103 / well, 1,2,4,8μmol·L-1Bdmc and 0.4,0.8,1.6,3.2μmol·L-1ADR respectively. The combined effects of 1 ~ 8μmol·L-1Bdmc and 0.8 ~ 3.2μmol·L-1ADR combined with HL-60 / ADR cells could produce simple synergistic effect. Sequential administration method, give SDR result after giving Bdmc synergisticly first, give Bdmc experimental result to give antagonistic effect after giving ADR first. Conclusion: Bdmc and ADR can produce synergistic effect at the same time. The sequential combination of Bdmc and ADR produces only one-way synergistic effect.
