目的 探索CYPⅡB2在正常与肝纤维化大鼠肝组织中的表达 ,评价螺内脂的治疗作用。方法 取雄性Wistar大鼠 16 0只 ,随机分为 4组 ,每组 40只。模型组 :CCl4 油 0 .2 5ml/ 10 0mg皮下注射 ,每周 3次 ;螺内脂组 :CCl4 油注射的同时予以螺内脂 2 0mg·kg-1·d-1灌胃 ,1次 /d ;马洛替酯组 :CCl4油注射的同时予以马洛替酯 5 0mg·kg-1·d-1灌胃 ,1次 /d ;对照组 :橄榄油皮下注射。光镜及电镜下动态观察组织学改变 ,图像分析仪测量胶原面积。RT PCR和原位杂交检测CYPⅡB2的表达。结果 原位杂交及RT PCR显示CYPⅡB2表达定位于星形细胞胞浆 ,在纤维化形成时表达增强。第 4、6周 ,螺内脂组肝纤维化分级、胶原面积低于模型组 (P <0 .0 5 )。第 8、10周 ,螺内脂组与模型组相比差异无显著性 (P >0 .0 5 )。结论 CYPⅡB2在纤维化肝脏的星形细胞中表达增强。螺内脂对早中期肝纤维化具有一定的治疗作用
Objective To investigate the expression of CYP Ⅱ B2 in the liver of normal and hepatic fibrosis rats and evaluate the therapeutic effect of spironolactone. Methods Sixty male Wistar rats were randomly divided into 4 groups (n = 40). Model group: CCl4 oil 0.52 / 10mg subcutaneously, 3 times a week; spironolactone group: CCl4 oil injection at the same time spironolactone 20mg · kg-1 · d-1 gavage, 1 / d; Mallottide group: CCl4 oil injection while malototer 50mg · kg-1 · d-1 gavage, 1 / d; control group: olive oil subcutaneous injection. Histological changes were observed under light microscope and electron microscope. The area of collagen was measured by image analyzer. RT PCR and in situ hybridization detection of CYP Ⅱ B2 expression. Results In situ hybridization and RT-PCR showed that CYPⅡB2 expression localized in the cytoplasm of astrocytes and was enhanced in the formation of fibrosis. At the 4th and 6th weeks, the hepatic fibrosis grade and collagen area in spironolactone group were lower than those in model group (P <0.05). At the 8th and 10th weeks, there was no significant difference between Spironolactone group and model group (P> 0.05). Conclusion CYP Ⅱ B2 is expressed in the astrocytes of fibrotic liver. Spironolactone on early and middle liver fibrosis has a certain therapeutic effect
