日的 :探讨顺铂 (CDDP)体内诱导肝细胞性肝癌 (HCC)细胞凋亡的发生情况及其临床意义。方法 :18例HCC患者随机分成 2组 ,10例行CDDP诱导治疗 ,另 8例作对照。用流式细胞仪 (FCM )检测 2组患者手术切除之肿瘤组织 ,作凋亡及细胞周期分析。结果 :CDDP治疗组肝癌细胞凋亡率较对照组显著增高 (P <0 .0 1)。CDDP治疗组肿瘤细胞G0 -G1 期较对照组明显增加 (P <0 .0 5 ) ;而S期细胞有显著减少 (P <0 .0 1)。细胞增殖指数 (PI)在CDDP治疗组显著降低 (P <0 .0 1)。结论 :CDDP在体内能显著增加HCC细胞的凋亡。小剂量CDDP诱导治疗为肝癌辅助治疗可行方法之一
Japan’s: To investigate the occurrence and clinical significance of apoptosis induced by cisplatin (CDDP) in hepatocellular carcinoma (HCC) in vivo. Methods: Eighteen patients with HCC were randomly divided into two groups. 10 patients received CDDP induction therapy and 8 patients received control. Flow cytometry (FCM) was used to detect the tumor tissues that were surgically removed in 2 groups for apoptosis and cell cycle analysis. Results: The apoptosis rate of hepatoma cells in CDDP treatment group was significantly higher than that in the control group (P < 0.01). In the CDDP-treated group, the G0-G1 phase of the tumor cells was significantly increased compared with the control group (P < 0.05), while the S-phase cells were significantly decreased (P <0.01). The cell proliferation index (PI) was significantly lower in the CDDP treated group (P < 0.01). Conclusion: CDDP can significantly increase the apoptosis of HCC cells in vivo. Low-dose CDDP induction therapy is one of the feasible methods for adjuvant treatment of liver cancer
