目的探讨整合素 β1在人脑胶质瘤细胞粘附和侵袭过程中的作用。方法用免疫组化SABC法 ,检测整合素 β1在人脑胶质瘤细胞SHG44中的表达 ;用细胞粘附实验检测其在SHG44细胞粘附中的作用。结果整合素β1可在人脑胶质瘤细胞SHG44中表达 ,抗整合素β1抗体可明显抑制人脑胶质瘤细胞SHG44对胶原蛋白I和胶原蛋白IV细胞外基质蛋白的粘附。结论整合素β1在人脑胶质瘤细胞的粘附和侵袭过程中必不可少 ,抑制其在人脑胶质瘤细胞中的表达,有助于阻止胶质瘤细胞的侵袭性生长。
Objective To investigate the role of integrin β1 in the adhesion and invasion of human glioma cells. Methods Immunohistochemical SABC method was used to detect the expression of integrin β1 in human glioma SHG44 cells. Cell adhesion assay was used to detect the role of integrin β1 in SHG44 cell adhesion. Results Integrin β1 was expressed in human glioma cells SHG44. Anti-integrin β1 antibody significantly inhibited the adhesion of human glioma SHG44 to collagen I and collagen IV extracellular matrix proteins. Conclusion Integrin β1 is essential in the adhesion and invasion of human glioma cells, inhibiting the expression of integrin β1 in human glioma cells and preventing the invasive growth of glioma cells.
