目的:应用液体蛋白芯片飞行时间质谱系统分析卵巢癌的血浆蛋白质表达谱,寻找具有潜在诊断意义的血浆肿瘤标志物,以利于卵巢癌的早期诊断。方法:收集卵巢浆液性癌30例、卵巢浆液性瘤18例、妇科良性疾病(包括卵巢囊肿和子宫肌瘤)30例的血浆样本,经磁珠纯化、基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)分析及ClinproTools生物信息学方法研究其血浆蛋白表达谱。结果:在卵巢癌组和非癌组间,得到差异性蛋白48个(P<0.05),用其中四个联合分组能力最强的蛋白质4 210.91(m/z),4 470.17(m/z),2 210.9(m/z),1866.88(m/z)建立了诊断模型,癌组和非癌组的识别率为100%。结论:利用液体芯片-飞行质谱技术进行血浆卵巢癌相关蛋白质的研究,得到了差异蛋白和诊断模型,对卵巢癌的诊断具有潜在价值。
OBJECTIVE: To analyze plasma protein profile of ovarian cancer using liquid protein chip time-of-flight mass spectrometry system and search for potential tumor markers for diagnosis of ovarian cancer. Methods: Thirty cases of ovarian serous carcinoma, 18 cases of ovarian serous tumors and 30 cases of benign gynecologic diseases (including ovarian cyst and uterine myoma) were collected and purified by magnetic beads. Matrix-assisted laser desorption / ionization time of flight mass spectrometry (MALDI -TOF-MS) analysis and ClinproTools bioinformatics method to study its plasma protein expression profile. RESULTS: Forty-eight differentially expressed proteins were found between the ovarian cancer group and the non-cancerous group (P <0.05). Among them, 4 210.91 (m / z) and 4 470.17 , 2 210.9 (m / z) and 1866.88 (m / z). The recognition rate of the cancer group and non-cancer group was 100%. Conclusion: The study of plasma proteins related to ovarian cancer using liquid chip-flight mass spectrometry has obtained differential proteins and diagnostic models, which have potential value in the diagnosis of ovarian cancer.
