目的 :研究脑星形细胞肿瘤中端粒酶活性与 S期细胞比例的相关性 ,探讨恶性肿瘤细胞周期变化对端粒酶活性表达的影响。方法 :采用端粒重复序列扩增方案 (TRAP) -银染法检测了 32例脑星形细胞肿瘤标本的端粒酶活性 ,并应用流式细胞术 (FCM)测定了它们的 S期细胞比例 (SPF)。结果 :在星形细胞瘤和恶性星形细胞瘤中 ,端粒酶活性的表达与 SPF无明显相关性 ;多形性胶质母细胞瘤端粒酶阳性标本的 SPF值明显高于阴性标本 ,经统计学分析得出两者差异有显著性。结论 :在细胞周期中端粒酶活性的改变可能受多种生物学因素的调控 ,其中恶性肿瘤本身的增殖特性可能起着极为重要的作用
Objective: To study the correlation between telomerase activity and the proportion of S-phase cells in astrocytic tumors and to investigate the effect of cell cycle changes on telomerase activity. Methods: The telomerase activity of 32 astrocytic tumor samples was detected by telomeric repeat amplification protocol (TRAP) - silver staining. The proportion of S phase cells was determined by flow cytometry (FCM) (SPF). Results: In astrocytoma and malignant astrocytomas, the expression of telomerase activity was not significantly correlated with SPF. The telomerase SPF value of glioblastoma multiforme was significantly higher than that of negative samples, The statistical analysis shows that there is significant difference between the two. CONCLUSION: The changes of telomerase activity in the cell cycle may be regulated by many biological factors. The proliferation of malignant tumors may play an important role
