目的:研究化瘀解毒方提取物抗胃癌细胞增殖和黏附方面的作用及机制。方法:采用高效液相色谱法(HPLC)分析化瘀解毒方提取物稳定性,化瘀解毒方提取物作用于人胃癌SGC-7901细胞株,利用MTT分析细胞增殖抑制实验,检测红细胞毒性实验,MTT快速测定法测定活性峰与Matrigel的黏附作用的影响。Western blot法检测人胃癌SGC-7901细胞中PI3K,PDK1和Akt总蛋白及其磷酸化水平的影响。结果:化瘀解毒方提取物可浓度依赖性抑制体外人胃癌SGC-7901细胞增殖、黏附,免疫印迹结果显示,化瘀解毒方提取物抑制Akt蛋白磷酸化,然而对于上游蛋白PDK1和PI3K的磷酸化无作用。结论:化瘀解毒方提取物抑制其增殖和黏附,其作用机制可能与抑制PI3K/Akt通路有关。
Objective: To study the effect and mechanism of Huayu Jiedu Prescription against gastric cancer cell proliferation and adhesion. Methods: The stability of Huayu Jiedu decoction was analyzed by HPLC. Huayu Jiedu decoction extract was applied to human gastric cancer cell line SGC-7901. MTT assay was used to analyze the cell proliferation inhibition test, the erythrocyte cytotoxicity test, Effect of Matrigel on Adhesion between MTT Assay and MTT Assay. Western blot was used to detect the total protein and phosphorylation of PI3K, PDK1 and Akt in human gastric cancer cell line SGC-7901. Results: Huayu Jiedu Fang extract could inhibit the proliferation and adhesion of human gastric cancer cell SGC-7901 in a concentration-dependent manner. Western Blot showed that Huayu Jiedu Fang extract inhibited the phosphorylation of Akt protein. However, for the phosphorylation of upstream protein PDK1 and PI3K No effect. Conclusion: Huayu Jiedu Fang extract can inhibit its proliferation and adhesion, the mechanism may be related to the inhibition of PI3K / Akt pathway.
